Abstract: Keratins (Ks) are the members of the family of the intermediate filaments expressed in the
epithelial tissues. Adult hepatocytes are unique, in that they express only K8/K18 whereas other cell
types display a more complex keratin expression pattern. The exquisitely simple keratin expression
pattern observed in hepatocytes is likely responsible for the fact that hepatocytes are highly
susceptible to keratin network perturbations. To illustrate this point, my presentation focuses on
three distinct aspects of keratin biology in human liver disease: (i) Inherited keratin variants
predispose to development and adverse outcome of both acute and chronic human liver disease; (ii)
keratins are redistributed into aggregates termed Mallory-Denk-bodies in a variety of chronic
disorders including alcoholic and non-alcoholic steatohepatitis (ASH,NASH); (iii) keratins are released
into the serum and represent promising markers of the extent of liver injury.
